Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

Effect of nutrition on Crassostrea gigas larval development and the evolution of physiological indices Part B: Effects of temporary food deprivation ArchiMer
Ben Kheder, Rym; Quere, Claudie; Moal, Jeanne; Robert, Rene.
In the natural environment bivalve larvae are exposed to variable conditions and can therefore face periods of food shortage. To understand the resistance of early life stages of Pacific oyster (Crassostrea gigas), larvae were experimentally starved for 4 days at different periods of their development, and the resulting variations in their lipid reserves then analysed using image analysis and biochemical techniques. Faced with a temporary lack of food, the larvae halted their growth in size and weight and started to live off their lipid reserves; this effect was highly marked when the period of food deprivation was at the beginning of larval rearing, but was less marked when food deprivation was after day 14. The larvae conserved their developmental...

Trophic niche of two co-occurring ophiuroid species in impacted coastal systems, derived from fatty acid and stable isotope analyses ArchiMer
Blanchet-aurigny, Aline; Dubois, Stanislas; Quere, Claudie; Guillou, Monique; Pernet, Fabrice.
The trophic niches of 2 common co-occurring ophiuroids, Ophiocomina nigra and Ophiothrix fragilis (Echinodermata, Ophiuroidea), in 2 contrasting coastal systems of Brittany (France) were investigated. We used a combination of fatty acid biomarkers derived from neutral lipids and stable isotopic compositions to explore the contributions of oceanic versus continental inputs to the ophiuroids' diet. We investigated 2 different systems with an inshore versus offshore comparison. We sampled potential food sources and surveyed organisms every 2 mo for 1 yr. Spatio-temporal variations in stable isotopes and fatty acid profiles of the ophiuroids were generally low compared to interspecific differences. Fatty acid markers showed that both ophiuroids relied on...

Assessment of oocyte and trochophore quality in Pacific oyster, Crassostrea gigas ArchiMer
Boulais, Myrina; Corporeau, Charlotte; Huvet, Arnaud; Bernard, Ismael; Quere, Claudie; Quillien, Virgile; Fabioux, Caroline; Suquet, Marc.
Pacific oyster hatchery production remains highly variable, partially due to inconsistent gamete quality. However, little attention has been paid to oocyte quality in Pacific oyster compared to important vertebrate aquaculture species such as fishes. The aims of the present study were to identify criteria of oocyte and trochophore quality used as predictive tools for embryo developmental success at the D-larval stage. Twenty-six potential proxies of oocyte and trochophore quality were studied on 25 mature females, as determined using histology. Some morphological and biochemical characteristics of mature oocytes were described. Protein appeared to be the main constituent of fully mature Pacific oyster oocytes. Neutral lipids accounted for 61% of the total...

Involvement of Mitochondrial Activity and OXPHOS in ATP Synthesis During the Motility Phase of Spermatozoa in the Pacific Oyster, Crassostrea gigas ArchiMer
Boulais, Myrina; Soudant, Philippe; Le Goic, Nelly; Quere, Claudie; Boudry, Pierre; Suquet, Marc.
In the Pacific oyster, spermatozoa are characterized by a remarkably long movement phase (i.e., over 24 h) sustained by a capacity to maintain intracellular ATP level. To gain information on oxidative phosphorylation (OXPHOS) functionality during the motility phase of Pacific oyster spermatozoa, we studied 1) changes in spermatozoal mitochondrial activity, that is, mitochondrial membrane potential (MMP), and intracellular ATP content in relation to motion parameters and 2) the involvement of OXPHOS for spermatozoal movement using carbonyl cyanide m-chlorophenyl hydrazone (CCCP). The percentage of motile spermatozoa decreased over a 24 h movement period. MMP increased steadily during the first 9 h of the movement phase and was subsequently maintained at a...

ATP content and viability of spermatozoa drive variability of fertilization success in the Pacific oyster ( Crassostrea gigas ) ArchiMer
Boulais, Myrina; Soudant, Philippe; Le Goic, Nelly; Quere, Claudie; Boudry, Pierre; Suquet, Marc.
Fertilization of the oocyte is a critical process of sexual reproduction depending among other factors on spermatozoa functionality. Mitochondria participate in many crucial processes for spermatozoa motility and fertilizing ability. The Pacific oyster, Crassostrea gigas, is characterized by a high inter-individual variability of its reproductive success which has been shown to be partially due to the variability of gamete quality. The present study explored spermatozoa characteristics such as: i) relationships among mitochondrial functionality (ATP content, mitochondrial membrane potential (MMP), reactive oxygen species production); sperm viability (percentages of live, dying, and dead); and motility; and ii) their involvement in inter-oyster variability...

Proteomic signatures of the oyster metabolic response to herpesvirus OsHV-1 μVar infection ArchiMer
Corporeau, Charlotte; Tamayo, David; Pernet, Fabrice; Quere, Claudie; Madec, Stephanie.
Pacific oyster Crassostrea gigas were inoculated with OsHV-1 at low load (control) or high load (challenged) to better understand the pathogenesis of ostreid herpesvirus 1 (OsHV-1 μVar) and to determine which metabolic pathways might be affected during infection. Animals were sampled for proteomic analysis two days post-injection, at the same time as OsHV-1 initiated an intense replication phase in challenged oysters. Twenty-five abundant protein spots that showed a marked change in accumulated levels were identified using a two-dimensional electrophoresis (2-DE) proteomic approach. Overall, these proteins are involved in cytoskeleton organization, protein turnover, induction of stress signals, signalling pathways and energy metabolism. Challenged oysters...

Proteomic identification of quality factors for oocytes in the Pacific oyster Crassostrea gigas ArchiMer
Corporeau, Charlotte; Vanderplancke, Gwenaelle; Boulais, Myrina; Suquet, Marc; Quere, Claudie; Boudry, Pierre; Huvet, Arnaud; Madec, Stephanie.
We used a 2-DE proteomic approach to identify abundant proteins linked to oocyte quality in the Pacific oyster Crassostrea gigas, an economically important bivalve. Oocyte quality of 14 females was estimated by recording fertilisation and early developmental success until D-larval stage under controlled conditions. Proteins that were differentially expressed between females showing high or low oocyte quality were identified by nano-liquid chromatography tandem mass spectrometry. Twelve up-accumulated spots associated with low quality oocytes revealed 10 distinct proteins, including vitellogenin — breakdown products and metabolic enzymes. Eight up-accumulated spots from high quality oocytes revealed 6 distinct proteins, including chaperone molecules and...

Influence of one selected Tisochrysis lutea strain rich in lipids on Crassostrea gigas larval development and biochemical composition ArchiMer
Da Costa, F.; Petton, Bruno; Mingant, Christian; Bougaran, Gael; Rouxel, Catherine; Quere, Claudie; Wikfors, G. H.; Soudant, P.; Robert, Rene.
Effects of a remarkably high overall lipid Tisochrysis lutea strain (T+) upon gross biochemical composition, fatty acid (FA), sterol and lipid class composition of Crassostrea gigas larvae were evaluated and compared with a normal strain of Tisochrysis lutea (T) and the diatom Chaetoceros neogracile (Cg). In a first experiment, the influence of different single diets (T, T+ and Cg) and a bispecific diet (TCg) was studied, whereas, effects of monospecific diets (T and T+) and bispecific diets (TCg and T+Cg) were evaluated in a second experiment. The strain T+ was very rich in triglycerides (TAG: 93–95% of total neutral lipids), saturated FA (45%), monounsaturated FA (31–33%) and total fatty acids (4.0–4.7 pg cell−1). Larval oyster survival and growth rate...

Standard and mutant Tisochrysis lutea strains were grown in batch culture for 10 days under nitrogen (N)-replete and N-reduced conditions to determine the effects of N supply in culture medium and growth phase on microalgal physiology and biochemistry. These two T. lutea strains were compared in terms of growth, morphology, associated free-living bacterial community, viability, intracellular lipid content (as measured by Bodipy staining), chlorophyll autofluorescence, and biochemical composition, with a focus on lipid class and fatty acid compositions. The standard strain (T) reached higher cell counts regardless of N supply in culture medium. In both T. lutea strains, microalgal final cell density was significantly lower in N-reduced medium. Carbohydrates...

Essential Fatty Acid Assimilation and Synthesis in Larvae of the Bivalve Crassostrea gigas ArchiMer
Da Costa Gonzalez, Fiz; Robert, Rene; Quere, Claudie; Wikfors, Gary H.; Soudant, Philippe.
Essential fatty acids (EFA) are important for bivalve larval survival and growth. The purpose of this study was to quantitatively assess for the first time through a mass-balance approach dietary EFA incorporation and synthesis within Crassostrea gigas larvae. A first experiment was carried out using two microalgae, Tisochrysis lutea (T) and Chaetoceros neogracile (Cg), as mono- and bi-specific diets. A second experiment using a similar design was performed to confirm and extend the results obtained in the first. Flow-through larval rearing was used for accurate control of food supply and measurement of ingestion. Non-methylene-interrupted fatty acids were synthetized from precursors supplied in the diet: 16:1n-7 and 18:1n-9, mediated by Delta 5...

Effect of a mono-specific algal diet on immune functions in two bivalve species - Crassostrea gigas and Ruditapes philippinarum ArchiMer
Delaporte, Maryse; Soudant, Philippe; Moal, Jeanne; Lambert, Christophe; Quere, Claudie; Miner, Philippe; Choquet, Gwenaelle; Paillard, Christine; Samain, Jean-francois.
The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. Oysters and clams were fed each of three cultured algae: Chaetoceros calcitrans, which is rich in 20:5(n-3) and 20:4(n-6) and poor in 22:6(n-3) fatty acids; T-Iso (Isochrysis sp.), which is rich in 22:6(n-3) and deficient in 20:5(n-3) and 20:4(n-6); and Tetraselmis suecica, which is deficient in 22:6(n-3) and contains only small amounts of 20:5(n-3) and 20:4(n-6). Fatty acid composition of haemocyte polar lipids was greatly affected by the diet. Oysters and clams fed C. calcitrans maintained a higher proportion of 20:5(n-3) and 20:4(n-6) in their haemocyte...

The Voltage-Dependent Anion Channel (VDAC) of Pacific Oysters Crassostrea gigas Is Upaccumulated During Infection by the Ostreid Herpesvirus-1 (OsHV-1): an Indicator of the Warburg Effect ArchiMer
Delisle, Lizenn; Fuhrmann, Marine; Quere, Claudie; Pauletto, Marianna; Pichereau, Vianney; Pernet, Fabrice; Corporeau, Charlotte.
Voltage-dependent anion channel (VDAC) is a key mitochondrial protein. VDAC drives cellular energy metabolism by controlling the influx and efflux of metabolites and ions through the mitochondrial membrane, playing a role in its permeabilization. This protein exerts a pivotal role during the white spot syndrome virus (WSSV) infection in shrimp, through its involvement in a particular metabolism that plays in favor of the virus, the Warburg effect. The Warburg effect corresponds to an atypical metabolic shift toward an aerobic glycolysis that provides energy for rapid cell division and resistance to apoptosis. In the Pacific oyster Crassostrea gigas, the Warburg effect occurs during infection by Ostreid herpesvirus (OsHV-1). At present, the role of VDAC in...

Laboratory conditioning modifies properties of gills mitochondria from the Pacific oyster Crassostrea gigas ArchiMer
Dudognon, Tony; Guderley, Helga; Quere, Claudie; Soudant, Philippe; Racotta, Ilie; Kraffe, Edouard.
Although laboratory experiments allow greater control of environmental conditions than field studies, they have several drawbacks. To analyze physiological responses to forcing environmental variables, experimental conditions should mimic natural conditions as closely as possible. For filter-feeding organisms in particular, diet quality and quantity is one of the environmental parameters that can differ markedly between experimental and field conditions. In the hatchery, Pacific oysters, Crassostrea gigas, commonly show good physiological performance and growth on a mixed algal diet of Tisochrysis lutea, formerly Isochrysis aff. galbana clone Tahiti (T-Iso), and Chaetoceros calcitrans, presumably as it provides a good supply of essential polyunsaturated...

Functional capacities of gill mitochondria in oyster Crassostrea gigas during an emersion/immersion tidal cycle ArchiMer
Dudognon, Tony; Soudant, Philippe; Seguineau, Catherine; Quere, Claudie; Auffret, Michel; Kraffe, Edouard.
Sessile animals that live on the foreshore undergo tidal cycles, and have to face variations in physical and chemical parameters such as oxygen concentration. During emersion, availability of dissolved oxygen can be lowered for bivalves, which have only a small reserve of seawater inside their closed shell. Differences in oxygen concentration are thus expected to lead to modifications of the metabolism, including changes in mitochondrial activity. Previous studies investigated air exposure under extreme conditions, which do not always reflect environmental conditions these invertebrates have to cope with. In this study, oxidative capacities of gill mitochondria of the oyster Crassostrea gigas were studied during a tidal cycle period, by comparing oysters...

Energy and Antioxidant Responses of Pacific Oyster Exposed to Trace Levels of Pesticides ArchiMer
Epelboin, Yanouk; Quere, Claudie; Pernet, Fabrice; Pichereau, Vianney; Corporeau, Charlotte.
Here, we assess the physiological effects induced by environmental concentrations of pesticides in Pacific oyster Crassostrea gigas. Oysters were exposed for 14 d to trace levels of metconazole (0.2 and 2 mu g/L), isoproturon (0.1 and 1 mu g/L), or both in a mixture (0.2 and 0.1 mu g/L, respectively). Exposure to trace levels of pesticides had no effect on the filtration rate, growth, and energy reserves of oysters. However, oysters exposed to metconazole and isoproturon showed an overactivation of the sensing-kinase AMP-activated protein kinase alpha (AMPK alpha), a key enzyme involved in energy metabolism and more particularly glycolysis. In the meantime, these exposed oysters showed a decrease in hexokinase and pyruvate kinase activities, whereas 2-DE...

Low pH reduced survival of the oyster Crassostrea gigas exposed to the Ostreid herpesvirus 1 by altering the metabolic response of the host ArchiMer
Fuhrmann, Marine; Richard, Gaëlle; Quere, Claudie; Petton, Bruno; Pernet, Fabrice.
Environmental change in the marine realm has been accompanied by emerging diseases as new pathogens evolve to take advantage of hosts weakened by environmental stress. Here we investigated how an exposure to reduced seawater pH influenced the response of the oyster Crassostrea gigas to an infection by the Ostreid herpesvirus type I (OsHV-1). Oysters were acclimated at pH 8.1 or pH 7.8 and then exposed to OsHV-1. Their survival was monitored and oyster tissues were sampled for biochemical analyses. The survival of oysters exposed to OsHV-1 at pH 7.8 was lower (33.5%) than that of their counterparts at pH 8.1 (44.8%) whereas levels of OsHV-1 DNA were similar. Energetic reserves, fatty acid composition and prostaglandin levels in oyster did not vary...
